Does wireless charging still work when it is on?

I had issues with wireless charging. I have a a Belkin and OnePlus stand charger and a Belkin flat desktop charger. Wireless charging was hit and miss with all three when using the case. Without the case, wireless charging works perfectly. I think because the back of the case stands away from directly touching the back of the phone, it's that which causes issues. I stopped using the case for that reason. Incidentally, prior to using the Spigen case I was using the official NOTHING case and had the same issues with that.

Can I use this case without the backplate - i.e. just the bumper around the edge?

no it's a full case with full back (but clear). I would highly recommend it though :)
