How long will one bottle last? How much do you use per cleaning?

The machine has a line that marks how much you should use. Keep in mind that this product is for the easy cleaning bissells. I often use more than recommended and it still lasts me, I would say about 15-20 uses (refills) maybe, I haven't tried to measure how long it lasts to be honest. I just know it works great for my needs, I have a Dog that is prone to accidents when he gets sick, now that he's old. I also moved into an apartment with a horribly dirty and sticky carpet, this product really did the trick, it turned that carpet back to its original color after a few washes (and if used with the pink carpet renewal it helps with the texture too) so to me, this is a necessity; but if you just need light cleaning, there are cheaper products that smell pretty and last longer. This has more of a fresh neutral smell that doesn't linger.

Can i use this with my bissell rug cleaner that does not have easy fill system? just mix it in with the water tank??

No. BISSELL Pet Stain & Odor + Antibacterial Formula controls odor-causing bacteria on upholstery and carpet. It works in all of our upright carpet cleaners that have an Easy Fill System. We formulated it to remove tough pet stains and be safe to use around kids and pets when it's used as directed.
