What are they maid of and how much can they hold if I have to lift with them?

They are made of Metal. If you use 2 handles to carry at the same time, they can hold up to 100 pounds.

What screws do you recommend? ( this should come with screws)

When it comes to the screws needed for your Speaker Cabinet Handles, we have you covered! Our screws are specially designed to provide the most secure and reliable fit, so you can rest easy knowing your cabinet handles are securely in place. Plus, this product comes with the screws you need included, so you don't have to go searching for them separately.

Will be installing in a pocket hole. if i remove the padding, how deep should i route the hole? thanks in advance for your response.

If you're planning to install the Speaker Cabinet Handles in a pocket hole, then you'll need to make sure the hole you route is at least 16mm deep. To get an accurate measurement, you might want to consider removing the padding in order to get the exact depth that you need.

Are there mounting screws included?

No they didn’t. Buy a bag of #8 self tapping wood screws. Also, the speakers I used them on were 100lbs a piece so I also used 4 machine screws with fender washers and lock-nuts with the nylon ring so the MDF would hold up better.
