Does it work with propylene or ethylene glycol?

It does work with propylene glycol. I do not see why it would be much different with ethylene glycol. I cannot be sure if the seals are at all damaged by the fluid. It is sad that beamnova is so difficult to find and ask a question. The first one I had was sold by Geindus, exact same unit visibly; and I believe but cannot be sure that the internals are all the same. It only lasted a year with only occasional use.

Can you intiially pump up to 5mpa then release pressure down to 3mpa?

I am not sure why you are using megapascals. Anyhow 725 psi I believe is at the high end of the pressure for this pump. The pump works like any hydraulic system - once you bleed fluid off some fluid pressure drops quickly. You can probably "catch it" at 3 mpa on its way down.
