Can I use this with sizzix machine?

you can if it is a die cutting machine

The description states this includes “1 set.” does this set include both pieces...the gate and pocket dies (2 pcs) , or just the gate portion (1 pc)?

You have to read listing, I order the flap only but they do have a set with flap and pocket die.

I make 4.25 x 5.5 cards will this die fit it?

The die measures 5 7/8". You could use it to make a 4.25x5.5 card...some of the design will be cut off (closest to the "stitched line near the fold on the item picture). But, it seems like it should still work...you will just have a small gap with unclosed details. I didn't get a chance to use this to actually create a 4.25x5.5 card yet.
